Inhalt:
We are going to talk about experimental design in the context of the analysis of neural circuits in health and disease. More specifically, we aim to study the cause of a devastating neurological disease, amyotrophic lateral sclerosis (ALS). One hypothesis is that development and progression of ALS is linked to neuronal excitability.
We are going to use the fruit fly Drosophila as an experimental model to explore this hypothesis. To this end, we have used Crispr/Cas9-based genome editing to "humagenize" the fly genome and introduce a human gene linked to ALS, and its mutant alleles. We now want to characterize this animal model, and will together discuss how to design a scientific study, plan specific experiments, and execute in the laboratory. We will suggest a general framework for this course and provide a set of tools and materials to approach the research questions from various angles. The students are asked to themselves develop the specific content of the course based on what they work out to be the most relevant or meaningful question. We will then use genetics and optogenetics, behavioral analysis, imaging for the physiological and anatomical analysis of nervous system function, as well as data analysis and statistics, to address the question specified.
